Legumes consumption is a usual and beneficial part of the human diet and contributing to health. Moreover, it is presently taking place a re-evaluation for its useful effects of consumption in the diet, which is the basis for various health claims. They are essential source of protein, carbohydrates, dietary fibre, micronutrients and various phytochemicals. Legumes have appreciable quantity of all the essential amino acids excluding sulphur containing amino acids, which can be balanced to combine with cereals in daily intake. Starch is the major stored carbohydrate followed by dietary fibre, simple sugars and oligosaccharides. Calcium, magnesium, potassium, phosphorus and iron are also present in legume seeds. Bioavailability of nutrients can be increased by soaking, sprouting and fermentation. Consumption of legumes reduces the risk of cardiovascular disease, some cancers (colon, breast and prostate) and also helps to manage body weight due to its satiety value. FAO has chosen 2016 as the International Year of Pulses which will emphasize the health and environmental benefits of pulses. Nutritional composition, anti-nutritional factors and health benefits of legumes are summarised in the current review.

Proteins are one of the basic and essential nutrients consumed by man. Both animal and vegetable sources provide proteins in the human diet. Many proteins derived from animal sources are nutritionally adequate because all essential amino acids are present. However, proteins from vegetable sources are often deficient in one or more essential amino acids. Therefore, vegetable protein must generally be supplemented with other proteins in order to provide good nutition. However, vegetable proteins can provide a satisfactory diet if the individual is judicious in selecting foods.A study of 1955 data indicated that approximately 55 percent of the protein component of the American diet came from animal sources. A recent analysis indicated that red meats contributed approximately 30 percent of the animal source to total protein. Dairy products, poultry, and fish contributed the balance. Similarly, wheat flour provided approximately 15 percent of total vegetable protein intake with other cereals (rice, corn, barley and rye), legume seeds (beans, peas and soybeans), and nuts constituting the balance.

Three digestibility experiments were conducted to evaluate the potential of determination of apparent ileal digestibilities (AID) of crude protein (CP) and amino acids (AA) by the mobile nylon bag technique (MNBT) using 21 feedstuffs and three mixed diets. In two conventional digestibility experiments (Exp. 1 and 2), AID were determined using in total 10 barrows (BW 35 kg) fitted with simple T-cannulas at the terminal ileum. For the MNBT studies, four pigs were fitted with a simple T-cannula at the proximal duodenum and a Post-Valve T-Caecum (PVTC) cannula at the terminal ileum. The MNBT studies included the feedstuffs (n = 10) from Exp. 1 and 2 as well as 14 further feedstuffs and mixed diets in which AID coefficients had been determined in previous trials. For each feedstuff 60 nylon bags were used. In vitro digestion of the bags was carried out in pepsin-HCl solution with 450 IU pepsin/l at pH 2.0 and 37°C for 4 h. In the 28-day in vivo experiment, 15 nylon bags per pig and day were inserted through the duodenal cannula and collected through the PVTC cannula after passage through the small intestine. Coefficients of AID were calculated based on the disappearance of CP and AA from the nylon bags during the in vitro and in vivo phase. In comparison with AID determined by the conventional method, AID of CP was on average 2.4% lower, whereas AID of lysine was on average 8.5% higher when determined by the MNBT. There was no significant (P > 0.05) correlation between AID coefficients of CP and AA determined by the conventional method and the MNBT, when all feedstuffs were taken into account. However, in cereals (n = 11), the correlation between AID coefficients determined by both methods was significant (P < 0.05) for CP (r = 0.61) and some AA (r ranging between 0.62 and 0.72). In conclusion, the potential of the MNBT to determine AID of CP and AA is rather limited. Differences in coefficients of AID of CP and AA were attributed to several factors such as diffusion of sample particles or endogenous protein through the nylon bags as well as to the presence of anti-nutritional factors (e.g. in legume seeds and oilseed meals).

For human nutrition the main source of vegetable proteins are cereal and legume seeds. The content of total soluble amino acids in mature endosperm of wild-type, opaque and floury maize (Zea mays L.) mutants were determined by HPLC. The total absolute concentration of soluble amino acids among the mutants varied depending on the mutant. The o11 and o13 mutants exhibited the highest average content, whereas o10, fl3 and fl1 exhibited the lowest average content. In general, the mutants exhibited similar concentrations of total soluble amino acids when compared to the wild-type lines, with the clear exception of mutants o11 and fl1, with the o11 mutant exhibiting a higher concentration of total soluble amino acids when compared to its wild-type counterpart W22 and the fl1 mutant a lower concentration when compared to its wild-type counterpart Oh43. For methionine, the mutants o2 and o11 and wild-type Oh43 exhibited the highest concentrations of this amino acid. Significant differences were not observed between mutants for other amino acids such as lysine and threonine. The high lysine concentrations obtained originally for these mutants may be due to the amino acids incorporated into storage proteins, but not those present in the soluble form.

AbstractThe most characteristic hallmark of life is its homochirality: all biomolecules are usually of one hand, e.g. on Earth life uses only L-amino acids for protein synthesis and not their D mirror images. We therefore suggest that a search for extra-terrestrial life can be approached as a Search for Extra- Terrestrial Homochirality (SETH). The natural choice for a SETH instrument is optical rotation, and we describe a novel miniaturized space polarimeter, called the SETH Cigar, which could be used to detect optical rotation as the homochiral signature of life on other planets. Moving parts are avoided by replacing the normal rotating polarizer by multiple fixed polarizers at different angles as in the eye of the bee. We believe that homochirality may be found in the subsurface layers on Mars as a relic of extinct life, and on other solar system bodies as a sign of advanced pre-biotic chemistry. We discuss the chiral GC-MS planned for the Roland lander of the Rosetta mission to a comet and conclude with theories of the physical origin of homochirality.

AbstractHydrogen cyanide polymers – heterogeneous solids ranging in color from yellow to orange to brown to black – may be among the organic macromolecules most readily formed within the Solar System. The non-volatile black crust of comet Halley, for example, as well as the extensive orangebrown streaks in the atmosphere of Jupiter, might consist largely of such polymers synthesized from HCN formed by photolysis of methane and ammonia, the color observed depending on the concentration of HCN involved. Laboratory studies of these ubiquitous compounds point to the presence of polyamidine structures synthesized directly from hydrogen cyanide. These would be converted by water to polypeptides which can be further hydrolyzed to α-amino acids. Black polymers and multimers with conjugated ladder structures derived from HCN could also be formed and might well be the source of the many nitrogen heterocycles, adenine included, observed after pyrolysis. The dark brown color arising from the impacts of comet P/Shoemaker-Levy 9 on Jupiter might therefore be mainly caused by the presence of HCN polymers, whether originally present, deposited by the impactor or synthesized directly from HCN. Spectroscopic detection of these predicted macromolecules and their hydrolytic and pyrolytic by-products would strengthen significantly the hypothesis that cyanide polymerization is a preferred pathway for prebiotic and extraterrestrial chemistry.

Fibers composed of different types of collagen cannot be differentiated by conventional electron microscopic stains. We are developing staining procedures aimed at identifying collagen fibers of different types.Pt(Gly-L-Met)Cl binds specifically to sulfur-containing amino acids. Different collagens have methionine (met) residues at somewhat different positions. A good correspondence has been reported between known met positions and Pt(GLM) bands in rat Type I SLS (collagen aggregates in which molecules lie adjacent to each other in exact register). We have confirmed this relationship in Type III collagen SLS (Fig. 1).
